1、实用医学杂志2014年第30卷第9期1373沉默中介体19对人乳腺癌MCF一7细胞侵袭及迁移的影响王晓艳郭子健吴小红李莉华摘要目的：研究中介体19(Medl9)对人乳腺癌MCF．7细胞侵袭及迁移的影响。方法：用慢病毒介导的Medl9小分子干扰RNA(siRNA)感染人乳腺癌细胞MCF一7(KD组)，并设置空载体转染~-(NC组)、空白对照组(CON组)。荧光显微镜下观察细胞感染效率，利用RT—PCR和Westernblot检测Medl9蛋白表达．用划痕实验和transwell实验检测细胞迁移及侵袭能

2、力。结果：荧光显微镜下观察细胞感染慢病毒效率达90％以上与NC组及CON组细胞相比，KD组Medl9mRNA表达水平下降了72_3％、72．1％，Medl9蛋白表达水平下降了85．4％、85_3％，差异均有统计学意义(P<0．05)。分别在0、6、12、24h比较迁移的距离，KD组细胞随着时间的延长，较CON组、NC组迁移能力显著下降，差异有统计学意义(P<0．05)；KD组、NC组、CON组MCF．7细胞穿过聚碳酸酯膜的细胞数分别为23．8±4．32、43．4±3．65、45．8±5．81．KD组
