3、metricassay.Theexpressionsofproliferationandapoptosisassociatedproteinsweredeterminedbyimmunocytochemicalmethod.RESULTS:AtimedependentanddosedependentproliferationinhibitionwasdemonstratedinTaoerqiextract.WiththeTaoerqiextracttreatmentof1,2and3mg/Lfor72h,theinhibit
4、oryratewas43.0%,54.9%,78.5%respectively.Thecharacteristicmorphologicalchangesofapoptosiswereobservedincellsaftertreatedby2mg/LofTaoerqi,andmanymultinucleategiantcellswerealsofound・TaoerqiinducedaG2/Mcellcyclearrestandtheapoptosiswastimeanddosedependent.Withthetreat
5、mentof2mg/LTaoerqifor48h,theproteinexpressionsofPCNAandBcl2decreasedfrom(154.78±0.16)and(85.56±0.09)to(7&46±0」5)and(40.43±0.07)(P0.01),whilep21WAFl/CIPlandcMycincreasedfrom(92.32±0.12)and(114.48±0.14)to(125.54±0.09)and(156.52±0.08)(P0.01).CONCLUSION:Thetimedependen
6、tanddosedependentproliferationinhibitoryeffectofTaoerqiextractonbreastcancercellsappearstobecausedbytheupregulationofp21WAF1/CIP1expression,downregulationofPCNAexpression,andtheapoptosisisrelatedtothedownregulationofantiapoptoticproteinBcl2andtheupregulationofcMycp
7、rotein.G2/Marrestisalsoassociatedwithapoptosis.[Keywords]Taoerqi;breastneoplasm;humanbreastcarcinomacelllinesMCF7;apoptosis【摘要】目的:研究桃儿七对人乳腺癌MCF7细胞增殖抑制和促进凋亡的作用,探讨其抗癌作用的机制.方法:应用MTT法检测桃儿七提取物对人乳腺癌MCF7细胞的生长抑制作用,倒置显微镜、HE染色及电镜观察细胞形态学改变,FCM分析细胞周期及凋亡率,免疫细胞化学法检测细胞增殖凋亡调控相关蛋白的
8、表达.结果淞儿七提取物可明显抑制MCF7细胞的增殖,1,2,3mg/L桃儿七作用72h后,细胞生长抑制率分别为43.0%,54.9%,78.5%,抑制作用呈时间及浓度依赖性.形态学观察到用药后凋亡细胞典型的形态学特征,伴有多核细胞形成.FCM分析发现桃儿七阻断MCF7细胞生长于细胞周期的G
