1、Rho/ROCK信号通路在腹膜透析大鼠腹膜纤维化中的作用及法舒地尔的干预效应孟绪初1甘平2(通讯作者)(1贵州医科大学研究生学院贵州贵阳550004)(2贵州医科大学附属乌当医院肾内科贵州贵阳550018)【摘要】目的:探讨Rho/ROCK信号通路在腹膜透析大鼠腹膜纤维化屮的可能机制。方法:将30只雄性SD大鼠随机分为四组:对照组,每日腹腔注射生理盐水25ml;模型组,每口腹腔注射4.25%的腹透液25ml;Fasudil组,每口腹腔注射含法舒地尔(Fasudil)浓度为10mg/Kg?d的4.25%腹透液2
2、5ml。实验第28天,取壁层腹膜组织行HE染色,并测定壁层腹膜ROCK-1的表达情况及SOD、MDA含量。结果:与模型组比较,Fasudil组不同程度腹膜厚度减少、胶原纤维沉积减轻,腹膜组织ROCK-1表达减少(P<0.05),MDA含量降低(P<0.05),SOD活性增加(P<0.05)。结论:腹膜透析液激活丫腹膜透析人鼠腹膜Rho/ROCK信号通路,促进腹膜组织氧化应激损伤,导致丫腹膜纤维化的发生,ROCK抑制剂法舒地尔能通过抑制此通路减少腹膜的氧化应激反应,进而延缓腹膜纤维化进展。【关
