4、ö;CTGF;ÊÓÍøĤ;ÌÇÄò²¡25¡¡¡¡Abstract¤rAIM:TodetecttheeffectofCTGFontheapoptosisinthediabeticretinawithsmallinterferingRNAs(siRNA)targetingwithCTGF.¤rMETHODS:Atotalof60ratsweredividedintosixgroupsincludingcontrolgroup,diabetic4,8,12,16weeksgroup,andinterferencegroup
5、.DiabeticratswereinducedbySTZintraª²peritoneal.At4,8,12,16weeksafterdiabeticsettingup,retinaswereobtainedfromcontrol,diabeticratsanddiabeticanimalstreatedbyintravitrealinjectionofCTGFsiRNAtosuppresstheexpressionofCTGFmRNA.RetinalcellsapoptosiswasdetectedbyTunnels
6、tainingandmRNAexpressionofCTGFwasanalyzedbyRTª²PCR.RESULTS:ThelevelsofCTGFandtheapoptosisintheretinasofdiabeticratsweresignificantlyhigherthanthoseinthecontrols.Apoptosisoccurredat4weeksafteradiabeticmodelsettingup,becameseriouswiththediabetesdeveloping,whileCTGF
7、elevatedat8weeks.Thecellapoptosiscountsincreasedto25.8cells/mm2at24weeksofdiabetes.SiRNAª²mediatedinhibitionofCTGFmRNAresultedinasignificantdecreaseinapoptosis.SignificantcorrelationswerefoundbetweenCTGFandapoptosisintheretina.¤rCONCLUSION:Theseresultssuggestthat
8、CTGFmightbeinvolvedinretinalcellsapoptosiswhichisacharacteristic25ofearlydiabeticretina.siRNAtargetingCTGFseemstohavetheadvantageofamelioratingretinalcellslost
