4、ract¤rAIM:TodetecttheeffectofCTGFontheapoptosisinthediabeticretinawithsmallinterferingRNAs(siRNA)targetingwithCTGF.¤rMETHODS:Atotalof60ratsweredividedintosixgroupsincludingcontrolgroup,diabetic4,8,12,16weeksgroup,andinterferencegroup.DiabeticratswereinducedbySTZintraª²peri

5、toneal.At4,8,12,16weeksafterdiabeticsettingup,retinaswereobtainedfromcontrol,diabeticratsanddiabeticanimalstreatedbyintravitrealinjectionofCTGFsiRNAtosuppresstheexpressionofCTGFmRNA.RetinalcellsapoptosiswasdetectedbyTunnelstainingandmRNAexpressionofCTGFwasanalyzedbyRTª²PCR

6、.RESULTS:ThelevelsofCTGFandtheapoptosisintheretinasofdiabeticratsweresignificantlyhigherthanthoseinthecontrols.Apoptosisoccurredat4weeksafteradiabeticmodelsettingup,becameseriouswiththediabetesdeveloping,whileCTGFelevatedat8weeks.Thecellapoptosiscountsincreasedto25.8cells/

7、mm2at24weeksofdiabetes.SiRNAª²mediatedinhibitionofCTGFmRNAresultedinasignificantdecreaseinapoptosis.SignificantcorrelationswerefoundbetweenCTGFandapoptosisintheretina.¤rCONCLUSION:TheseresultssuggestthatCTGFmightbeinvolvedinretinalcellsapoptosiswhichisacharacteristic25ofea
