1、产漆酶裂褶菌筛选和其产酶培养条件优化探究 摘要:从采自秦岭的森林腐木菌苔上分离到一株产漆酶的裂褶菌(SchizophyllumcommuneFr.)菌株mys005,对其产漆酶培养方式、培养基组成及培养条件进行了优化试验。结果表明,裂褶菌菌株mys005的产漆酶适宜培养方式为液体浅层振荡培养,含有Cu2+、Mn2+、Zn2+的微量元素复合液、洋葱(AlliumcepaL.)、麦(TriticumaestivumL.)麸对其产漆酶都有较大的促进作用,而愈创木酚则对其生长和产漆酶有抑制作用。由新鲜马铃薯(SolanumtuberosumL.)200g、K2HPO4
2、1.00g、NaCl0.50g、MgSO4·7H2O0.50g、NaNO32.50g、CaCl2·2H2O0.10g、FeCl30.02g、吐温-801.20mL溶于1000mL去离子水中组成的培养基在添加微量元素复合液(50mg/mLCuCl2·2H2O+20mg/mLMnSO4·H2O+10mg/mLZnSO4·7H2O)3mL、洋葱100g、麦麸10g后,并且培养基起始pH4.5、培养温度30℃、培养摇床转速200r/min、培养时间7d,则摇瓶发酵的漆酶产酶量最高可达1121U/mL,显示出了良好的工业开发潜力。关键词:漆酶;裂褶菌(Schizophyl
