1、遵竖医堂院亟±堂僮i金塞主窒擅要N一乙酰半胱氨酸对大鼠肺纤维化模型中结缔组织生长因子表达影响的研究中文摘要目的:探讨N一乙酰半胱氨酸(N—acetylcysteine,NAC)对博莱霉素(bleomycin,BLM)诱导的大鼠肺纤维化模型的肺组织结缔组织生长因子(connectivetissuegrowthfactor,CTGF)的蛋白和基因表达的变化及其意义。通过对用药前后肺组织病理学改变的观察及治疗前后肺组织结缔组织生长因子蛋白和基因表达变化的研究,探讨NAC对肺纤维化的作用及其可能的作用机制,为NAC应用
2、于临床提供理论依据。方法:取健康、雌性Wistar大鼠30只,随机分为BLM组(B组),NAC治疗组(N组)及正常对照组(C组),每组10只。B组及N组通过气管内注入BLM法制备肺纤维化模型。C组在同样条件下向气管内注射等量的生理盐水,N组于BLM处理前一周及此后每日经强饲法口服NAC(3mmol/kg)。三组动物分别于气管灌注后第7天、第14天随机处死5只,取大鼠的部分肺组织进行病理检测、免疫组化测定肺组织中CTGF的蛋白表达;通过RT-PCR检测剩余肺组织CTGFmRNA的表达变化。结果:病理学观察发现B组
