1、VEGF反义寡核苷酸对淋巴瘤新生血管生成的抑制作用临床血液学杂志2007年1且筮鲞蔓塑VEGF反义寡核苷酸对淋巴瘤新生血管生成的抑制作用童允洁邹萍张敏郭荣.?37?[摘要]目的:研究硫代磷酸化修饰的血管内皮生长因子(VEGF)反义寡核苷酸(ASODN)对裸鼠淋巴瘤血管生成的作用.方法:将终浓度为20~mol/L的VEGFASODN,错义序列及PBS分别与人淋巴瘤细胞系Namal—wa细胞孵育24h,裸鼠随机分为VEGFASODN组,错义序列组和对照组,将上述处理的Namalwa细胞(5X10溶于PBS液
2、2O1)接种于棵鼠皮下,4周后处死全部裸鼠,测定肿瘤大小,标本采用链酶菌抗生素蚤白一过氧化酶免疫组化法(SP法)检测肿瘤组织微血管密度(MVD).结果:第28天VEGFASODN组,错义序列组和PBS组肿瘤体积的大小分别为(212.6土196.5),(468.2土161.3)和(473.1土166.7)mm.VEGFASODN组微血管密度(12.26土0.78)较对照组(24.13土l_21)显着减少.结论:VEGFASODN对裸鼠淋巴瘤的生长及其新生血管的生成有明显的抑制作用.[关键词]Namalwa
