1、.640.安徽医药AnhuiMedicalandPharmaceuticalJournal2015Apr;19(4)没药甾酮对胆管癌RBE细胞移植瘤生长和miRNA表达谱的影响仲飞,童铸廷,范璐璐,姚梦群,孙国平(安徽医科大学第一附属医院,安徽合肥230022)摘要:目的研究没药甾酮对胆管癌RBE细胞株移植瘤生长和miRNA表达的影响。方法雌性SD大鼠建立胆管癌RBE细胞株移植瘤模型,随机分为对照组和没药甾酮高、低剂量实验组;高、低剂量实验组大鼠分别予40、20mg·kg~·d没药甾酮灌胃,对照组
2、大鼠予生理盐水灌胃。每5天测量裸鼠质量和肿瘤体积,处理30d后取材,提取肿瘤组织总RNA并分离miR—NA。采用微阵列杂交,通过芯片扫描和数据分析获得miRNAs表达谱。结果高、低剂量实验组大鼠移植瘤体积分别在处理20d和25d后显著低于对照组(P<0.01);处理30d后高、低剂量组移植瘤质量均显著低于对照组(P<0.01)。芯片结果显示,高剂量实验组瘤组织miRNAs表达谱与对照组相比有显著差异。结论没药甾酮可以显著抑制胆管癌RBE细胞株移植瘤生长并影响其miRNA的表达。关键词:没药甾酮;胆
