1、摘要结核分枝杆菌是典型的胞内寄生菌,主要靶细胞是巨噬细胞,但树突状细胞能显著激活初始型T细胞,启动T细胞免疫应答反应,是细胞免疫反应的重要启动者。MicroRNAs作为一类18—22nt的小分子,其调控作用广泛存在真核生物的各个进程,而且在免疫方面尤为重要,本研究采用miRNA芯片明晰BCG感染前后DCs中miRNA表达谱的变化,以发现可能参与DCs调控结核分枝杆菌感染相关miRNA,并采用RealtimePCR进行初步验证,为阐明DCs抗结核分枝杆菌的特异性miRNA调控机制奠定基础。1、成功分离
2、并培养出健康人外周血来源的树突状细胞,经过BCG感染DCs前后MicroRNA芯片检测,显示miRNA表达谱的差异,上调的有hsa-miR一1270;hsa.miR一147b;hsa-miR-3201;hsa-miR.132:hsa-miR.3611:hsa-miR.155;hsa-miR-1973;hsa-miR.934;hsa-miR一106b*等30个,下调的有hsa-miR一27b;hsa.miR-151-5p;hsa-miR一374c;hsa-miR一425;hsa-miR一125b;hs
