2、ratories,Lincoln,Nebraska,U.S.A.AcceptedSeptember20,1989Summary.Thecharacteristicsofatemperaturesensitivefelineinfectiousperi-tonitisvirus(TS-FIPV)wereexamined.TS-FIPV,unlikeitsparentstrain,DF2wildtypeFIPV(WT-FIPV),propagatedat31°C(permissivetemperatur
3、e)butnotat39°C(nonpermissivetemperature).ThistemperaturepreferenceofTS-FIPVwasalsodemonstratedincatsbytheabilityofthevirustoreplicateonlyatthelowertemperatureintheupperrespiratorytractandnotatsystemicsiteswherehighertemperatures(38-39°C)prevail.Viralst
4、ructuralproteinsandRNAweresynthesizedat39°CbutsomeundefinedmaturationaldefectpreventedtheformationofinfectiousTS-FIPVatitsnonpermissivetemperature.TS-FIPVwasmorethennolabilethanWT-FIPVwhichindicatedalterationsinthestructuralproteinsofTS-FIPV,andadiffer
5、enceintheenvelopeproteinofthetwoviruseswasrevealedbyWesternblotanalysis.PlaqueassaycharacterizationshowedthatTS-FIPVproducedsmallplaquesincomparisontothelargeplaquesofWT-FIPV.TheseuniquecharacteristicspossessedbyTS-FIPVmayaccountforitsnonvirulentnature
6、andabilitytostimulateprotectiveimmuneresponsesincats.IntroductionFelineinfectiousperitonitis(FIP)isacomplexandfataldiseaseofcatscausedbyinfectionwithfelineinfectiousperitonitisvirus(FIPV).Previousattemptstoconsistentlyprotectcatsbyimmunizationwithother
7、antigenicallyrelatedcoronavirusessuchasporcinetransmissiblegastroenteritisvirus(TGEV)[26],caninecoronavirus(CeV)[2],andhumancoronavirus[1]havebeenunsuc-cessful.InconsistentprotectionwasfoundwhencatsweregivenasublethaldoseofvirulentFIPVandcatsvaccinated
8、withanavirulentFIPVweremoreeasilyinfectedthanwerenonvaccinatedcats[14].Recently,ithasbeendem-onstratedthatanintranasally(IN)administeredtemperaturesensitiveFIP¥(TS-FIPV)vaccineisefficaciousandsafeuponFIPVchallenge[5].186K.K.Christianson
