1、哈尔滨工业大学工学硕士学位论文AbstractThefruitbodyofTricholomaMatsutakeisoneofthemostimportantediblefungis,whichisnotonlyverydeliciousandnutrimental,butalsorichinpolysaccharidesthathavemultiplepharmacologicalactivitiesandbioactivities.TheactivitiesofTricholomaMatsutakepolysaccharides(TMP)involveantitumor,enh
2、ancingimmunity,antiradiancyandsoon.Inthisstudy,theextraction,purification,modification,andantitumoractivityofTricholomaMatsutakefruitbodieswereinvestigated.Themainresultsareasfollows:Theoptimaloperationparametersforextractingwere:theratioofmaterialtowateris1:30,theextractiontemperatureis90℃,an
3、ddurationis3hoursrespectively.Extractiontwiceiseconomy(therateoftheextractionsisupto94.3%).PrecipitatedtheTMPwith3timesvolumeof95%ethanolfor6histhebestchoiceinthisstudy.TheproteininthepolysaccharidesremovedwiththeSevagmethod.Ionexchangeresinmethodwasthebestoneinallofactivecarbon,H2O2andtheresi
4、nfordecolorationbycomparingthecolorandpolysaccharides’lossrates.AfterthedialysiswiththedialysisbagwhoseMolecularweight(MW)isover8000,celluloseDE-52columnchromatographywhichelutedwithofwaterandNaClsolutionwasusedtoseparatefinepolysaccharides.TMWEP-1、TMWEP-2、TMWEP-3wereobtainedwhentheconcentrati
5、onofNaClsolutionwere0,0.2mol/Land0.5mol/L,respectively.TheTMWEP-2waspurifiedtoTMWEP-2ⅠandTMWEP-2ⅡbySephadexG-100gelcolumn.TheTMWEP-2ⅠwasidentifiedasahomogeneouscomponentbymethodsofSephadexG-100andHPLC.Itwaswhitesolidafterdialyzing,concentrating,anddesiccating.TMPwasmodifiedwithsnailenzymes.The
6、effectofmodificationmainlycomesfromfourfactors:duration,ratioofenzymetoTMP,pHandtemperature.Viathemono-factortestandorthogonaltest,weconcludedthattheoptimumconditionofmodificationweretheduration=5h,theratio=1:5,thepH=6.0andthetemperature=40℃.SGC-7901、Hela、HT-29werethreekindsoftumorcells.Theywe
