2、nesil一1.1中,构建出含靶基因片段的shRNA真核表达载体PGenesil—TGF一131一A、PGenesil—TGF—B1一B及PGenesil—HK,行酶切鉴定及测序分析,并采用脂质体介导法体外转染Hela细胞,荧光显微镜观察转染结果。经酶切和测序鉴定证实,构建的shRNA成功插入到载体,重组质粒的插入序列与设计的靶基因片段完全一致,3种重组质粒均能转染入Hela细胞表达绿色荧光。成功构建出靶向大鼠TGF—Bl基因的shRNA真核表达载体,为后续研究抗肝纤维化的基因药物奠定了实验基础。关键词:肝纤维化;TGF一131;RNA干扰;质粒;基因重组中图分类号:R318文献标识码:
