4、freemedium．FlowcytometrywasusedtodetecttheexpressionofCD133inHPACcellsandspherecells．TheefectsofhybridomamcAbsonself—renewal，proliferationandchemosensitivitytoGemcitabineofHPACparentorspherecellswereidentifiedbyserum—freesuspensioncultureandCCK一8assay．Results：HPACcellscouldsurvive．

5、proliferateandformspherecellsinserum—freemedium．Thesphere—formingratewas4．8％±0．6％．ThepercentageofCD133cellspopulationinspherecellsincreasedby11．6foldscomparedtoHPACcells．ThepositiverateofCD133inHPACspherecellswassignificantlyhigherthanthatinHPACparentcells．Amongthe20candidatemcAbs．
