2、共转染至包装细胞293T,包装产生病毒颗粒后感染Jurkat细胞,获得Syncytin稳定沉默的细胞系.荧光显微镜下观察感染效率,分别使用RealtimePCR和AnnexinV—APC单染法结合流式细胞仪检测目的基因表达水平和细胞凋亡率.结果成功构建了具有Syncytin干扰效果的慢病毒载体及其稳定沉默的Jurkat细胞系.结论该细胞系的构建为进一步研究Syncytin基因在白血病的功能提供了细胞模型.[关键词]RNA干扰;慢病毒载体;Syncytin基因;Jurkat细胞系[中图分类号]Q291[文献标识码]A[文章编号]2095-610X(2014)08一o012—04C0ns
