1、上海交通大学硕士学位论文乳腺特异表达人血小板生成素基因载体的研究姓名:刘茵申请学位级别:硕士专业:生物化学与分子生物学指导教师:黄英20070511乳腺特异表达人血小板生成素基因载体的研究达水平要明显高于BLG3.9和BLG1.9启动子调控的TPO真核表达载体。但随着时间推移,细胞数目不断地增加,pB3.9T却远远超过了pPT。第六,获得稳定整合了三种载体的单克隆成纤维细胞系。我们可以得出以下结论,第一,山羊β-乳球蛋白基因和β-酪蛋白启动子都能够指导TPO在乳腺细胞中特异表达。第二,在BLG基因5’端启动子上游-3.9kb至-1.9kb区域内可能含有特殊的增强序列。第三,随着细胞的不断增值

3、Humanthrombopoietin(TPO)istheall-importantregulatoryfactorwhichcanregulatethegenerationofthrombocyteinhumanbody.Itcanspecificallyimprovethelevelofthrombocyteinhumanbody,preventandcurethrombocytopeniacausedbychemotherapy,andalsopreventandcureidiopathicthrombocytopenicpurpura.Furthermore,thelatestres
4、earchshows,asacellregulatoryfactor,TPOcanbeusedtoacceleratethegrowthofhemopoieticstemcells.Theobjectiveandmeaningofthisresearchare:InordertoproducetransgenecloningcowwhichcanexpresshumanTPOhighefficientlyinitsmilk,eucaryonexpressedvectorspilotedbyvariouspromotersareconstructed,andspecificallyexpres
