3、ranalyzingthefactorsresponsiblefordifferingsterolcomposition,thefield·grownandculturedmyceliawereextractedwith80%ethanolatroomtemperatureandtotalsterolswerepreparedusingsilicongelcolumnchromatographyfollowedbyidentificationusingeitherGC—MSorspectroscopicmethods.ForculturingInonotusobllquus

4、,theseedculturewasgrowneitherinbasicmediumconsistingofglucose(2%),yeastextract(0.5%),KH2PO4(0.01%),MgSO4·7H2O(0.05%)anddistilledwateratpH6.5,orthebasicmediumsupplementedwithserialconcentrationsofAgNO3.Theresultsindicatedthatfield—grownmyceliacontainedlaflosterolandinotodiol(comprised45.47%

5、and25.36%ofthetotalsterols。respectively)andother10sterols(comprisingtheremaining30.17%)includingergosterolbiosyntheticintermediatessuchas24·methylenedihydrolanosterol,4,4-dimethylfecosterol,4-methylfecosterol,fecosterolandepistero1.Columnchromatographyalsoledtotheisolationoflanosterol,Inot

6、odiol,trametenolicacid,foscoparianolBandanewtriterpenoidfoscoparianolDinfield-grownmycelia.Incomparison,theculturedmyceliaonlycontainedthreesterolswithergosterolasthepredominantone(82.20%).Lanosterolonlyaccountedfor3.68%.SupplementingAgintothecuhureat0.28Ixmol·Lgreatlyenhancedcontentoflano

7、sterol(accountingfor56.81%)anddecreasedthecontentofergosterol(18.5%)togetherwiththepresenceofintermediatesforergosterolbiosynthesis.Theseresultssuggestedthatthesterolcompositioninmyceliaofthefunguscanbediversifiedbysupplementingsubstancesinhibitingenzymaticpro
