1、r曲人学硕l“学位论文酿酒酵母3一磷酸甘油脱氢酶基因和3一磷酸甘油酯酶基因在大肠杆菌中的共表达摘要作为改造代谢途径获得直接转化葡萄糖生产l,3一丙二醇的基因工程菌的第一步,本论文对产甘油基因工程菌的构建做了以下研究。首先,从酿酒酵母Saccharomycescerevisiae中克隆3.磷酸甘油脱氢酶(GPDl)和3·磷酸甘油酯酶(HOR2)基因,分别构建重组质粒pSE-gpdl和pSE—hot2,然后进行两种重组基因的表达试验,一种是双表达盒法,另一种是多顺反子法。在第一种表达法中含双表达盒的重组EcoliJMl09菌株在含l%葡萄糖的摇瓶发酵培养基中于37
2、"C培养24小时,有甘油产生,产甘油率为1.189,'L。在第二种表达方法中以JMl09为宿主的含多顺反子的重组菌株在含1%葡萄糖的摇瓶发酵培养基中于37*C培养24小时,有甘油产生,产甘油率为3.799/L。而以BL21作为宿主时,重组菌株发酵罐发酵28小时,产甘油率为22.649/L,葡萄糖的转化率为45.3%研究结果表明:将gpdl和hot2基因引入大肠杆菌,在大肠杆菌中构建了一条可将葡萄糖直接转化成甘油的新的代谢途径是可行的。同类研究在国内尚无报道。关键词:甘油3.磷酸甘油脱氢酶3.磷酸甘油酯酶大肠杆菌共表达发酵广州人学硕十学位论文Co-EXPRESS

6、sioncassetteswereinsertedintoplasmidpGEM.3zf(p)andtransformedintoEcoliJMl09.TherecombinantEcoliJMl09wasincubatedinMinimalmedia11%glucoseshake.flasksat37℃,withvigorousshakingfor24h,afterwhichtheyweresampledforHPLCofsupematant.TherecombinantEcoliJMl09showedproductionofglycerolWas1.189
