1、2型糖尿病肾损伤大鼠模型的建立和肾脏CD34表达作者:胡波,李锋,李敏,王胜春【摘要】目的观察高脂喂养联合低剂量链脲佐菌素(STZ)注射的SD大鼠2型糖尿病肾损伤模型的代谢特征及CD34的变化。方法4周龄雄性SD大鼠60只随机分为2组。正常对照组10只,普通饲料喂养;糖尿病模型组50只,高脂饲料喂养,即普通饲料中加入1O%猪油、10%葡萄糖、1%胆固醇、2%蛋黄粉、1%胆酸钠。喂养4周后尾静脉注射STZ(35mg/kg)。所有大鼠在STZ注射10d与30d时做灌胃葡萄糖耐量试验(OGTT)。放免法测定血清胰岛素,免
2、疫组化染色观察肾脏CD34的阳性表达。结果糖尿病肾损伤大鼠空腹血糖(FBG)显著高于正常对照组(P<0.01),血糖值在注射完STZ后1~3个月比较稳定;24h尿蛋白从1个月后一直持续上升;空腹血清三酰甘油(TG)和游离脂肪酸(FFA)水平显著高于正常对照组(P<0.05);尿素氮(BUN)、血肌酐(Cr)显著高于正常对照组(P<0.01);在糖尿病肾损伤大鼠的肾脏中CD34出现高表达。结论高脂喂养联合低剂量STZ注射的糖尿病大鼠模型拟2型糖尿病肾病发生的主要病理生理过程,具有高血糖以及血脂异常和

6、thanthatinthecontrolgroup(P<0.01),bloodglucosestayedstable1-3monthsafterinjectionofSTZ,whilethe24hurineproteinkeptrisingafter1month,fastingserumtriglyceride(TG)andfreefattyacids(FFA)weresignificantlyhigherthanthatincontrolgroup(P<0.05),withsignificantlyh
7、igherbloodureanitrogen(BUN)andserumcreatinine(P<0.01).CD34showedhighexpressionininjuredkidneyoftheratswithdiabetes.ConclusionThemainpathologicalandphysiologicalprocessoftheoccurrenceoftype2diabetesmodelratswithhigh4greasefeedingandlow-doseSTZinjectionembody
