1、基质金属蛋白酶3在创伤愈合中作用的实验研究世界急危重病医学杂志2007年3月第4卷第2期IntJEmergandCritCareMed.March..2007V01.4,N0.2?1751??实验研究?基质金属蛋白酶3在创伤愈合中作用的实验研究文亮屈纪富刘明华【摘要】目的探讨基质金属蛋白酶3在创伤愈合过程中的变化及其意义.方法取伤后1d伤口液作为创伤修复启动微环境,刺激真皮多能干细胞,24h后检测细胞表达MMP3含量的变化.同时制作单纯创伤和难愈创伤动物模型,通过免疫组化和图像分析方法观察伤后不同时相伤口组织MMP3含量变化.结果创伤早期伤口液作用真皮多能干细胞后表达MMP3显着增加.正常大鼠
2、皮肤组织成分可见一定量的MMP3表达,阳性细胞主要为表皮细胞,毛囊外根鞘细胞,真皮成纤维细胞,血管内皮细胞等.单纯创伤大鼠伤后各时相点MMP3表达均显着增加.特别是以真皮组织增加更为明显,其峰值在伤后57d.难愈创伤组大鼠伤口局部MMP3含量在伤后各时相点亦显着增加,但与单纯创伤组相比,其含量在伤后3,5,7d均显着减少,尤以真皮组织表达减少更为明显,并且MMP3表达峰值延迟,为伤后10d.结论MMP3是一种重要的创伤愈合调节分子;通过高表达MMP3可能是真皮多能干细胞参与创伤修复的途径之一.【关键词】创伤;愈合;基质金属蛋白酶3;难愈创伤;真皮多能干细胞中图分类号:R641文献标识码:A文章
