1、新型KATP开放剂iptakalim对人肺动脉平滑肌KATP通道mRNA表达的论文摘要】目的研究新型katp开放剂iptakalim对原代培养人肺动脉平滑肌细胞katp通道亚基sur2b/kir6.1mrna表达的影响,以探讨iptakalim治疗肺动脉高压的分子生物学机制。方法分离人手术标本正常肺组织3~4级肺小动脉,原代培养人肺动脉平滑肌细胞。正常组不予干预,试验组分别加入内皮素1(et1)、et1+不同浓度iptakalim或glyburide等孵育24h。以trizol法抽提总rna,逆转录为cdna,以实时荧
2、光定量逆转录聚合酶链反应(rqrtpcr)法检测各组细胞katp通道mrna表达量。结果et1组sur2bmrna的表达量较正常组明显减少。iptakalim呈浓度依赖性拮抗et1作用,提高sur2bmrna表达量。glyburide呈浓度依赖性拮抗iptakalim作用,减少sur2bmrna表达量。各组kir6.1mrna表达量无统计学差异。结论新型katp开放剂iptakalim呈浓度依赖性增加原代培养人肺动脉平滑肌细胞katp通道mrna表达量,可能成为较有前途治疗肺动脉高压的有效药物。【关键词】atp依赖性
