1、...页眉SDF-1在牵张成骨中的表达及作用机制研究[摘要]:目的探讨基质细胞衍生因子-1(SDF-1)在大鼠下颌骨中牵张成骨(distractionosteogenesis,DO)区的表达情况及其作用机制的初步分析。方法40只SD大鼠随机分为8组(每组5只),全部将右侧下颌骨表面皮肤切开,放置下颌骨牵张器,调整塑形并固定。而后将大鼠左侧下颌骨骨折行钛板内固定手术以作对照。随后分别于不同的时间点(0d,3d,6d,9d,12d,15d,18d,21d)麻醉处死,解剖大鼠双侧下颌骨后,小心刮取右侧牵张间隙骨痂及左侧骨折处骨痂进行免疫组织化学检测和酶联免疫吸附测定。结果在40只实
2、验大鼠中,伤口愈合正常,手术耐受性良好;免疫组化染色结果显示,SDF-1在成骨细胞和微血管周围表达较为明显,且在DO的新生骨痂中的表达较骨折处骨痂中的表达更加明显;酶联免疫吸附检测结果显示:SDF-1无论在DO区或是骨折区,其数值均增加3倍,而术后一周后,SDF-1在骨折区的表达情况呈下降趋势,而在SDF-1的表达情况在进入牵张期内其再次上升,高达原有水平的5倍,牵张结束后缓缓下降,且两组SDF-1的表达情况分别9d、12d、15d具有统计学差异(P<0.05)。结论SDF-1在牵张成骨区和骨折区的表达程度差异明显,为日后研究SDF-1在牵张成骨中的表达及作用机制奠定了基础。
