1、广州医学院硕士学位论文BDE-209对体外培养海马神经干细胞增殖分化及凋亡的影响阳性,GFAP阳性,DAPI阳性细胞数目,计算NF+/DAPI+,GFAP+/DAPI+比例,并测量NF+细胞突起长度。【结果】NF+细胞比例及NF+细胞突起长度随着BDE-209浓度的增加而减少,GFAP+细胞比例随着BDE-209浓度的增加而增加,存在剂量依赖关系.【结论】BDE-209可以导致神经干细胞分化成神经元的比例发生改变,随BDE-209浓度增加神经干细胞分化成神经元的比例有不同程度的下降,分化成神经元的突起长度也有不同程度的下降。【关键词】BDE-209;神经干细胞;分化第
2、四部分BDE-209对体外培养新生鼠海马神经干细胞凋亡的研究【目的】观察BDE-209对海马神经干细胞凋亡的影响。【材料与方法】1.传代培养3-4代的新生鼠海马神经干细胞暴露于BDE-209,实验共分5组,空白对照组,DMSO对照组,实验A组:BDE-209浓度为10ug/ml,实验B组:BDE-209浓度为30ug/ml,实验C组:BDE-209浓度为50ug/ml。每组设3个平行样。DMSO对照组加入含1‰DMSO的培养液。24h后进行检测。2.采用免疫细胞化学技术检测神经干细胞凋亡。【结果】不同浓度的BDE-209作用24h后,均可致神经干细胞出现凋亡。实验A,
3、B,C组作用24h后其凋亡率均有明显增高,表明BDE-209可导致海马神经干细胞凋亡,与对照组相比差异均有显著意义(P<0.01)。【结论】BDE-209在一定剂量和作用时间内可致海马神经干细胞的凋亡。推测BDE-209在一定浓度范围内可诱导神经干细胞凋亡,这可能是导致仔鼠学习记忆能力下降的作用机制之一。【关键词】BDE-209;凋亡;神经干细胞;6广州医学院硕士学位论文BDE-209对体外培养海马神经干细胞增殖分化及凋亡的影响RoleofBrominateddiphenlyethers-209inproliferation,differentiationandapo
