1、不同剂量rhBMP-2对兔下颌骨牵引成骨区新骨生成的影响及其OPG表达【摘要】目的研究不同浓度rhBMP-2在不同时间对兔下颌骨牵引成骨区新骨生成的影响及牵引区新骨中骨保护素(osteoprotegerin,OPG)的表达。方法在48只成年大耳白兔的一侧下颌骨前部行骨切开术,分别将空白胶原、rhBMP-21.5mg和rhBMP-23mg胶原复合物植入下颌骨切开处,用牵引器延长一侧下颌骨4mm,稳定期第1、3、7、14天,分别处死各组动物,取牵引区新生骨痂行OPG免疫组化染色。结果下颌牵引延长后牵引间隙均有新骨形成,免疫组化染色OPG主要定位于成骨细胞的胞浆中。
2、在同一时间内,应用rhBMP-2组较对照组有显著性差异(P<0.05),且rhBMP-23mg剂量组较rhBMP-21.5mg剂量组有显著性差异(P<0.05)。结论动物实验表明,rhBMP-2能促进兔下颌骨牵引成骨区新骨的生成并具有浓度依赖性。【关键词】牵引成骨rhBMP-2骨保护素 Abstract:ObjectiveToinvestigatetheEffectsofdifferentconcentrationsofrhBMP-2onmandibulardistractionosteogenesisandtheexpressionofOPGinthepe

5、gionaftercompletionofdistraction.Themandibularsidetreatedwith1.5mgrhBMP-2gainedgreateramountofnewboneformationthantreatedwithcollagensponge,andthemandibularsidetreatedwith3mgrhBMP-2harvestedgreateramountofnewboneformationthantreatedwith1.5mgrhBMP-2.OPGexpressionwasfoundinthecytoplas
6、mofosteoblastsandstainedbrownordarkbrownbyimmunohistochemistry.Inthemeantime,OPGexpressionindicatedtherewassiganificantdifferencebetweenrhBMP-2groupandcontrolgroup(P<0.05),and3mgrhBMP-2grouphadsiganificantdifferencecomparingwith1.5mgrhBMP-2group(P<0.05).ConclusionsTheanimalexperimen
