4、ignificantdifferenceingrainproteincontentwereusedtoinvestigatetheexpressionlevelandstructureofglutaminesynthetasegene(GS),activityofglutaminesynthetaseandproteinaccumulationcharacteristicsduringgrainfilling.Theresultsshowedthattransgressivevariantscouldbeobtainedthroughsuccessivedirectiveselection
5、ofgrainproteincontent,becausethegrainproteincontentwascloselyrelatedtocorrespondinggenotype.Duringgrainfilling,theactivityofglutaminesynthetasefollowedasingleGpeakcurve,highlycorrelatedwithgrainproteincontentandsignificantlydifferedbetweenparentsandprogenies.GS1.3andGS2genes,intransgressivevariant
6、sandtheirparentswithdifferentproteincontent,exhibitedsimilartranscriptiontrendduringgrainfilling,thatis,thetranscriptionlevelincreased15-20daysbeforeheadingandthendecreasedgradually.Moreover,thegrainproteincontentwascloselyrelatedtoGS1.3andGS2expressionlevels.ThevarietieswithhigherGStranscriptionl
7、evelshowedhigherproteincontentcomparedwiththeirparents.Inaddition,althoughGS1.3andGS2genesequencesshowedhighconservation,thegenesequenceandproteinsequenceofGS1.3andGS2werenotcompletelyidenticalindifferentvarietie
