3、,2016,30(4):345G355.Abstract:Brittleculmmutantsareoneoftheimportantmaterialstostudyplantmechanicalstrength.Abrittleculmmutantnamedasbc16(brittleculm16)wasisolatedfromNipponbarethroughEMS(EthylMethaneSulphonate)treatment.Comparedwithwildtype,bc16featuredfragileandeasilybrokenstems,
4、leaves,roots,lowerplantheightandfilledgrainsperpanicle,shorterpaniclelengthandmainrootlength.Thecomponentsanalysisofstemcellwallshowedthatthecellulosecontentinbc16mutantwassharplydecreased,whilehemiGcellulosecontentwasevidentlyinGcreasedandnosignificantdifferencehadbeenfoundinthel
5、ignincontentbetweenbc16mutantandwildtype.Thecellshapeandarrangementofparenchymacellsinbc16mutantbecameirregularanddisordered.Moreover,theparenchymacellwallsandthesecondarycellwallsofsclerenchymacellsinbc16mutantundertheepidermislayerwerethinnerthanthoseinwildtype.Theresultsofgenet
6、icanalysisrevealedthatthemutanttraitwascontrolledbyasinglerecessivegene,whichwasmappedina66.6GkbregionbetweenInDel(insertionGdeletion)markers2GFand2GHonchromosome2with7candidategenes.Amongthesegenes,Os02g0738900,whichisallelictoBC3gene,wasspeciallyfocusedon.thTherewasaTtoAsubstitu
7、tionattheveryendofthe13thintronofOs02g0738900whichwasatthe5113baselocaGtionfrominitiationcodonbysequencing,whichledansplicingforwardmutationintheprocessoftranscription,andsixnucleotideswerecutintomRNAasaresult,whichfinallycausethestopoftranslation.Additionally,inthemutant,therewer
8、esignificantdecreaseintheexpressi
