4、antwasobtainedfromtheF2populationofthreeGwaycrossⅡG32B//XieqingzaoB/DularandnamedasBrittleculm18(Bc18)accordingtoitsphenotypes.Eachpartofplantshowedbrittlenessduringthewholegrowthperiod.Inordertoidentifythemutant,nearisogeniclinepopulationsofZhongcuiBandZhong9Bwerecrea
5、tedwithBc18mutantasthedonorofbrittlegeneandZhong9B,anormalculmGstrengthvariety,asthereceptorandrecurrentparent.Comparedwithwildtype,Bc18significantlydeclinedby70.70%and47.16%inmechanicalstrengthofculmandleaves,respectively.Nosignificantdifferenceswerefoundintermsofgrow
6、thduration,plantheight,paniclenumberperplant,spikeletnumberperpanicle,seedsettingrateand1000Ggrainweight.CellwallcomponentsanalysesshowedthatcellulosecontentandlignincontentinBc18hadnosignificantdifferenceascomparedwiththoseofwildtype,whilehemicellulosecontentinculmand
7、leafdramaticallyincreasedby31.84%and17.35%,respectively.GeneticanalysesofsixF2andtwelveBC1F1backcrosspopulationsrevealedthatthephenotypeofBc18wascontrolledbyasingledominantgene.Bc18/02428andBc18/9311F2populationsweredevelopedforBc18genemapping.BymeansofmapGbasedcloning
8、technology,withsomeSSRmarkerspublishedonlineandnewdesignedInDelmarkers,Bc18waslocalizedbetweenInDelmarkerPBC22andPBC3
