5、································26中文摘要青皮中橙皮苷提取及纯度测定陈秀秀河北北方学院医学院药学系药学专业07级目的:通过优化文献提取橙皮苷的方法得到提取橙皮苷的最佳工艺条件并采用TCL法进行定性鉴别(2010版药典第一部);采用高效液相色谱法(HPLC)测定橙皮苷的纯度。方法:以青皮为原料,用碱液和甲醇做溶剂提取橙皮苷的条件为:粗苷:甲醇:碱液[NaOH浓度为0.1%]=1:23:47在85oC以上浸泡4h,调节pH值为4。即先将甲醇和碱液搅拌混合,再将粗苷加入,继续搅拌1h使粗苷溶解完全,然后过滤
6、至滤液澄清,用盐酸调滤液的pH值至4,静置沉淀,过滤,滤饼用煮沸过的水洗涤至无色,甲醇滤液精馏回收,橙皮苷的收率达85%以上。TCL鉴别出与橙皮苷对照品相对应的宽点;HPLC法测定出橙皮苷纯度为85.94%且理论塔板数不低于1000。结论:此方法操作简便,易控制且提取橙皮苷的效率较高,所建立的定性定量方法专属性强,重复性好。关键词:青皮;橙皮苷;薄层色谱法;紫外分光光度法;高效液相色谱法AbstractExtractionandpurificationofhesperidinincicusprocessingandmeasuringi
