3、loftheChineseAcademyofSciences,Beijing100039,ChinaABSTRACT.AlthoughtheperitrichousciliateCarchesiumpolypinumiscommoninfreshwater,itspopulationgeneticstructureislargelyunknown.Weusedinter-simplesequencerepeat(ISSR)fingerprintingtoanalyzethegeneticstructureof48differentisolatesofthespeciesfromfourlake
4、sinWuhan,centralChina.Usingeightpolymorphicprimers,81discernibleDNAfragmentsweredetected,amongwhich76(93.83%)werepolymorphic,indicatinghighgeneticdiversityattheisolatelevel.Further,Nei’sgenediversity(h)andShannon’sInformationindex(I)betweenthedifferentisolatesbothrevealedaremarkablegeneticdiversity
5、,higherthanpreviouslyin-dicatedbytheirmorphology.Atthesametime,substantialgeneflowwasfound.Sothemainfactorsresponsibleforthehighlevelofdiversitywithinpopulationsareprobablyduetoconjugation(sexualreproduction)andwidedistributionofswarmers.Analysisofmo-lecularvariance(AMOVA)showedthattherewaslowgeneti
6、cdifferentiationamongthefourpopulationsprobablyduetocommonancestryandfloodingevents.Theclusteranalysisandprincipalcomponentanalysis(PCA)suggestedthatgenotypesisolatedfromthesamelakedisplayedahighergeneticsimilaritythanthosefromdifferentlakes.BothanalysesseparatedC.polypinumisolatesintosub-groupsacco
7、rdingtothegeographicallocations.However,thereisonlyaweakpositivecorrelationbetweenthegeneticdistanceandgeographicaldistance,suggestingaminoreffectofgeographicaldistanceonthedistributionofgeneticdiversitybet
