3、要目的:通过对大鼠缺血缺氧性脑损伤模型注射不等含量的神经生长因子,并和正常大鼠模型的不同部位神经生长因子浓度和受体含量进行对比,得出神经生长因子和其受体在缺血缺氧性脑损伤中的作用机制,为临床指导治疗缺血缺氧性脑损伤提供依据。方法:1.大鼠模型资料:选取南方医科大学动物中心的16-18周大鼠若干只,雌雄不限,模型均随机筛选,统计结果无统计学差异。2.方法:将这些随机抽取的大鼠模型一共50只,按照是否手术和手术注射神经生长因子的情况的不同分为5组,A组的大鼠模型正常喂养,B组的大鼠模型做左颈动脉结扎手术,并且在持续喂养
4、时每日注射生理盐水;C组的大鼠模型做左颈动脉结扎手术,并且在持续喂养时每日注射鼠神经生长因子30ug;D组的大鼠模型做左颈动脉结扎手术,并且在持续喂养时每日注射鼠神经生长因子60ug;E组的大鼠模型做左颈动脉结扎手术,并且在持续喂养时每日注射鼠神经生长因子90ug,比较这五组大鼠模型在术后1天、术后一周、术后两周、术后四周这几个时间段头部、腹部和尾部不同部位神经生长因子浓度和TrkB受体含量的变化情况。3.数据分析:各组大鼠模型的基本资料情况经正态性检查,服从正态分布,且各组间无统计学差异。使用SAS6.12统计学
5、软件按照2*2析因设计进行统计学分析,分别通过对大鼠模型术后的统计指标结果做出评估。数值型变量以均数±标准差表示。结果:五组大鼠模型在术后相同时间内同组不同部位的神经生长因子浓度和TrkB受体含量的统计学差异具有统计学意义(P<0.05);在术后相同时间内不同组别同部位的神经生长因子浓度和TrkB受体含量的统计学差异具有统计学意义(P<0.05);在术后不同时间内同组别同部位的神经生长因子浓度和TrkB受体含量的统计学差异具有统计学意义(P<0.05)。关键词:神经生长因子;TrkB受体;缺血缺氧性脑损伤;作用机制
