1、2013年l2月第3O卷第12期ChinJExpSu.December2013.Vo1.30.No.12·2547··泌尿外科·严重联合免疫缺陷小鼠肾包膜下异种移植人前列腺增生组织模型的建立吴建辉杨阔杨宇明马洪顺徐勇【摘要】目的利用严重联合免疫缺陷(SCID)小鼠肾包膜下移植法建立人前列腺增生(BPH)组织异种移植模型。方法将20只4~6周龄SCID小鼠随机分为4组,每组5只。取前列腺术后组织标本,分别移植到小鼠肾包膜下和皮下。实验组为肾包膜下移植,对照组为皮下移植。8~l2周后,观察前列腺组织生长,经石蜡包埋后进行常规染色和免疫
2、组织化学鉴定。结果人BPH组织在SCID小鼠肾包膜下成活率为90%(18/20),皮下组仅为40%(8/20)。异种移植的前列腺组织为实体状、不规则、白色,隆出肾脏表面并嵌入肾实质,伴有血管生成。镜下表现,肾包膜下移植前列腺组织生长旺盛,皮下组前列腺组织腺体萎缩,移行上皮化生。免疫组织化学染色证实为人源性前列腺组织。结论小鼠肾包膜下移植可高效率建立人BPH组织异种移植动物模型。【关键词】前列腺增生;异种移植;睾酮;模型,动物Establishmentofanexperimentalmodelofhumanbenignprostat
