2、、胎牛血清培养基(胎牛血清组)、无血清培养基(无血清组)培养BMSCs,传代后换用胎牛血清培养;12、24、48、72h和5d首次换液:绘制P3代BM.SCs生长曲线;免疫荧光鉴定BMSCsCIM4和vimentin的表达;诱导BMSCs的骨向、内皮向分化;茜素红检测钙结节表达;免疫荧光鉴定血管内皮样细胞CD31和vwF的表达。结果(1)同种异体血清组及胎牛血清组细胞形态均一,生长速率相似。(2)P3代BMSCs生长曲线显示:同种异体血清组与胎牛血清组倍增速率相似,均快于无血清组。(3)P3代BMSCs免
3、疫荧光染色显示:同种异体血清组与胎牛血清组CD44、vimentin表达均高于无血清组。(4)成骨诱导14d后,茜素红染色可见大量橘红色钙化结节形成。(5)内皮诱导14d后,内皮样细胞表面标志物CD31、vWF阳性。结论改良培养条件后所获得的骨髓细胞具有问充质干细胞的表型和功能特点。【关键词】骨髓间充质干细胞;全骨髓贴壁;倍增时间;定向分化Modifiedcultureconditionsofbonemarrowmesenchymalstemcellsanddirectionaldiferentiatio
