1、环球中医药2013年8月第6卷第8期GlobalTraditionalChineseMedicine，August2013，Vo1．6，No．8589·论著·基于MTT法的波棱瓜子抗肝纤维化用量初步研究徐和戴领沈成英韩晋袁海龙【摘要】目的制定波棱瓜子质量控制标准含量上下限，以此确定波棱瓜子用于肝纤维化治疗的合理用量范围。方法本文采用中压制备色谱收集不含波棱甲素(herpetrione，PEDX)的阴性样品，加入不同剂量的PEDX，制备不同浓度梯度的PEDX系列样品溶液，建立PEDX系列样品HPLC指纹图谱，四甲基偶氮唑盐(thiazolyl

2、bluetetrazoliumbromide，MTI")比色法观察PEDX系列样品溶液对人肝星状细胞系LX一2细胞增殖的影响，并比较PEDX系列样品溶液与波棱瓜子提取物溶液的抑制率差异。结果(1)指纹图谱结果显示，不同浓度梯度的PEDX系列样品相似度良好；(2)PEDX系列样品能抑制LX一2细胞增殖，加入样品的剂量M与细胞抑制率I值呈现一定的“量一效”关系，确定了波棱瓜子中药效成分PEDX的最小有效用量为0．68mg，最大有效用量为8．2mg。结论波棱瓜子用于肝纤维化治疗的合理用量范围为3．27～39．42mg。【关键词】中药；波棱瓜子；波
