1、·3212·中国基层医药2013年11月第2O卷第21期ChinJPrimMedPharm，November2013，Vo1．20，No．21小鼠肝癌实验模型建立及生物治疗效果观察易永祥赵伟吴引伟魏强韩建波龙启强方媛苏舒【摘要】目的探讨建立小鼠肝癌原位模型并观察其生物治疗的效果。方法选取Balb／c小鼠来源的ML．3肝癌细胞经肝内直接注射接种至肝脏实质内，与经脾注射肝癌细胞相比较，对其成瘤情况进行观察；并予以生物治疗，观察其疗效。结果肿瘤细胞接种第6天起，经脾脏注射组以及经肝脏注射各组小鼠均不同程度出现体质量减轻、腹

2、水、黄疸以及腹腔可触及包快。在经脾脏注射组小鼠中，5只小鼠中仅有1只小鼠长出肝脏肿块。其余均腹腔种植转移。在经肝脏直接注射小鼠中，15只小鼠中有10只长出肝癌，腹腔中没有种植转移8例。DC肿瘤疫苗联合0X86单抗治疗组小鼠的全身状况要优于疫苗单独治疗组以及PBS对照组。分别选取对照组小鼠以及治疗组小鼠的脾脏细胞以及腹水细胞样本进行流式细胞分析，治疗组脾脏和腹水细胞中MDSCs，即表达CD11bLyC6细胞(2．77％)低于对照组(11．94％)(=5．36，P<0．05)。结论经肝脏直接注射肿瘤细胞较经脾脏注射法能更
