1、ScienceofSericulture_■冒鳖鼙一2014,4o(2):0202—0209ISSN0257—4799;CN32—1115/SE—mail:CYKE@chinajourna1.net.cn以多年生麻栎茎段为外植体的组织培养条件优化于艳庆龙夏兴宏孙娟王连珍高伟李立峰姜欣雨(辽宁省蚕业科学研究所,辽宁凤城118100)摘要麻栎是柞蚕的饲料树种之一。以5年生麻栎的茎段为材料,研究茎段消毒时间、培养基及其重要组分含量、暗培养时间等因素对外植体成活、不定芽诱导及幼苗生根的影响,优化麻栎茎段的组织培养技术条件。茎段外植体采用1LHgC
2、I,消毒5rain,其成活率最高达85.0%;在MS、WPM、1/4MS(2倍铁盐)3种基础培养基中外植体的不定芽诱导率均在82.5%以上,其中以I/4MS(2倍铁盐)培养基的诱导率最高(86.7%),且外植体的褐死率仅有5.0%;茎段外植体初代培养的暗培养时间以5—7d为宜。以1/4MS(2倍铁盐)为基础培养基,优化各时期培养基中植物生长剂的添加浓度:茎段外植体初代培养的最适培养基配方为1/4MS(2倍铁盐)+1.0mg/L6-BA+0.2mg/LIBA+1.0mg/LKT,在此培养基中不定芽的诱导率可达92.0%;不定芽继代培养基的最
3、适配方为1/4MS(2倍铁盐)+2.0mg/L6-BA+0.2mg/LIBA+1.0mg/LGA,在此培养基中不定芽生长健壮,增殖系数为5.8;幼苗生根培养基的最适配方为1/4MS(2倍铁盐)+1.0mg/LIBA,在此培养基中再生植株的生根率达74.0%。采用上述优化的组织培养技术条件,可有效提高多年生麻栎茎段的不定芽诱导率及再生植株成活率。关键词麻栎;组织培养;茎段;消毒时间;培养基配方;暗培养时间中图分类号$336;$889.3文献标识码B文章编号0257—4799(2014)02—0202—08OptimizationofTiss
