3、obiol.,20:131-142.Theporcineepidemiccoronavirus(PEDV),tentativelyclassifiedasacoronavirus,wasadaptedtoVerocellsandaplaquetestdevelopedforinfectivitytitration,allowingustotestthebiologicalandbiophysicalpropertiesofthevirus.Growthkineticsshowedpeaktitersofl0ss'plaq
4、ue-formingunitsml-~15hafterinfection.Filtrationexperimentsandelectronmicroscopyrevealedaparticlediameterbetween100and200nm.Thebuoyantdensityoftheviruswas1.18.Theparticlelostitsinfectivityontreatmentwithlipidsolvents.Virusreplicationcouldnotbeinhibitedby5-iodo-2'-
5、deoxyuridine.PEDVwasmoderatelystableat50°C,butheatsensitivitywasnotalteredbydivalentcations.At4°C,theviruswasstablebetweenpH5.0and9.0,butat37°CstabilitywasrestrictedtothepHrange6.5-7.5.Viralinfectivitywasnotimpairedbyultrasonicationorbymultiplefreezingandthawing.
6、PEDVwasnotneutralizedbytransmissiblegastroenteritisvirusantiserum.Onthebasisofthetestscarriedout,PEDVisapleomorphic,envelopedRNAviruswithaparticlediameterof~150nmandabuoyantdensityof1.18.Infectivitydependsonthepresenceoftrypsin,andinfectedcellsshowatendencytofuse
7、andtoformsyncytia.Alloftheseproperties,aswellasitsphysicochemicalcharacteristics,allowPEDVtobeclassifiedasacoronavirus.INTRODUCTIONTransmissiblegastroenteritisvirus(TGEV)andporcineepidemicdiarrheavirus(PEDV),thefirstatrueandthesecondaprobablememberofthecoron-avir
