2、ork1220l-0509ReceivedMay2,1990;acceptedJuly11,1990Toobtaininformationaboutthestructureandevolutionofthenucleocapsid(N)proteinofthecoronavirusmousehepatitisvirus(MHV),wedeterminedtheentirenucleotidesequencesoftheNgenesofMHV-A59,MHV-3,MHV-S,andMHV-1fromcDNAclones.Atthenucl
3、eotidelevel,theNgenesequencesoftheseviralstrains,andthatofMHV-JHM,weremorethan92%conservedoverall.Evenhighernucleotidesequenceidentitywasfoundinthe3’untranslatedregions(3’UTRs)ofthefivestrains,whichmayreflecttheroleofthe3’UTRinnegative-strandRNAsynthesis.AllfiveNgeneswer
4、efoundtoencodemarkedlybasicproteinsof454or455residueshavingatleast94%sequenceidentityinpairwisecomparisons.However,aminoacidsequencedivergenceswerefoundtobeclusteredintwoshortsegmentsofN,putativespacerregionsthat,together,constitutedonly11%ofthemolecule.Thus,thedatasugge
5、stthattheMHVNproteiniscomposedofthreehighlyconservedstructuraldomainsconnectedtoeachotherbyregionsthathavemuchlessconstraintontheiraminoacidsequences.Thefirsttwoconserveddomainscontainmostoftheexcessofbasicaminoacidresidues;bycontrast,thecarboxy-terminaldomainisacidic.Fi
