3、ccDNAprobes.BCV-specificrecom-binantplasmidprobesp52,p27,andp247,holdinginsertsderivedfrom(prob-ablynonstructural)genes,andplasmidspN17andpN9holdingtheNandMgene,respectively,permittedthedetectionofisolatesofbothBCVandTCVwithsimilarsensitivities.Similarly,p
4、robingsupernatantsofcellculturesinfectedwithseveralisolatesofTCV,usingprobespN17andpM78,respectivelyholdingtheNgeneofBCVandTCV,resultedinequallyintensedetectionsignals.OnlyaslightdetectionofMHV-3,whichisantigenicallyrelatedtoBCV,wasob-served,whereastheprob
5、esdidnotallowthedetectionofIBV,TGEV,andHCV-229E,whichareplacedinantigenicgroupsseparatefromthoseofBCVandTCV.DetectionofTCVwasimprovedbyhybridizationwithBCV-specificsingle-stranded(ss)probesholdingsequencesoftheNandMgenesandsyn-thesizedbythepolymerasechainr
6、eaction.DiagnosisofTCVin134clinicalsamplesbyhybridizationwasbetterwithPCR-producedssBCV-specificprobesthanwithdsPCR-producedprobesoracombinationofsixrecombinantplasmidprobesholdingnon-overlappingBCV-specificcDNAsequences.Detectionsig-nalswereabsentwhenprob
