3、inecoronavirus(BC)adaptedtotissueculture,wasinoculatedintoor-ganculturesofbovinefoetaltrachea.Haemagglutinininthefluidfrominfectedorganculturesreachedtitresof32andcharacteristiccoronavirusparticleswereobservedelec-tronmicroscopically.BCvirusantigenwasde
4、tectedinfrozensectionsoftheorgancul-turesbystainingwithfluorescentantibody.ThesedatawereevidencethatBCvirusre-plicatedinorganculturesofrespiratorytissue.Theuseofthistechniqueforprimaryiso-lationofbovinecoronavirusesfromfieldmaterialisdiscussed.INTRODUCT
5、IONMostcoronavirusessofardescribedhavetwosignificantbiologicalcharac-teristics.First,theyhaveamarkedtropismforeithertherespiratoryoren-terictractandsecondly,theyaredifficulttoisolateincellculture.Thebovinecoronavirus(BCvirus)isassociatedwithneonataldiar
6、rhoea(Stairetal.,1972),andthusresemblesthevirusoftransmissiblegastroente-ritis(TGE)ofswineandarecentlyreportedhumanvirus(CaulandClarke,1975).Incontrast,avianinfectiousbronchitisvirus(Cunningham,1966),theratcoronavirus(Parkeretal.,1970)andmostcoronavirus
7、esofman(Brad-burneandTyrrell,1971)areassociatedwithrespiratorydisease.However,recentreports(Underdahletal.,1974;Kemenyetal.,1975)indicatethatTGEvirusinfectionhasarespiratoryphase.Therefore,itispossiblethatBCvirusmightreplicateinrespiratorytissue.Thebovi
8、necoronaviruswasoriginallycharacterisedbyanimalpassageafterpurificationfromfaeces(Stairetal.,1972).Lateritwasgrownincellculturewithdifficulty(Mebusetal.,1973a).Sinceinoculationoforgancul-turesoffoetaltrachea(HoornandTyrrell,1969)
