3、s.IntroduetionRecentlyitwasestablishedthatthemucoidsurface-filmoflungalveolaecontainsaveryhighconcentrationofascorbieacid.Whiletherearedifficultiesindeterminingtheascorbicacidcontentofthesurfacefilm,itwasestimatedtobebetween1.0and3.5mg/ml(16).Becauseitwasthoughtthattheseconcentration

4、smaybehaveasadefensivebarrier,systemsusingmonolayerculturesofchick-embryorespiratoryepithelium,allantois-on-shellandchick-embryoeiliatedtrachealorgan(CETO)cultureswereusedtostudytheeffectofascorbicacidonvirusinfection.ThevirusesusedweremembersoftheFamiliesOrthomyxoviridae(influenzaA)

5、,Paramyxoviridae(Newcastlediseasevirus,NDV),andCorona-viridae(avianinfectiousbronchitisvirus,IBV).MaterialsandMethodsRespiratoryepitheliumcultureswerepreparedeitherin"Falcon"tissuecultureflasks(30ml)orin"Linbro"96cuppolystyrenetissueculturemicroplatesIS-FB-96-TC.Allantois-on-sbellpre

6、parationsweremadebythemethoddescribedbyFAZE~:ASand%V]c[ITE(6).CETOcultureswerepreparedbythemethodofCttER~¥andTAYLOR-ROBI~SO~(4)withminormodifications.Thetracheaweredissectedoutfrom20day-oldchick-embryosandcuttransverselyintorings1mmthick.Afterwashing,theringswereplacedindividuallyint
