2、SUMMARYGenomicRNAextractedfromthepurifiedmurinecoronavirusJHMsedimentsbetween52Sand54Sinaqueoussucrosegradients.TheRNAissingle-strandedandhasanapparentmol.wt.of5"4to6"5×~o6,asdeterminedbyelectrophoresisinpolyacrylamideagarosegelsofdifferentconcentrations.Thepresen
3、ceofpoly-adenylatesequencesintheRNAisdemonstratedbybindingtooligo-(dT)celluloseanddigestionwithribonucleasesAandTI.ThepurifiedRNAdoesnotdissociateintosubunitsathightemperaturesorinhighconcentrationsofDMSOandisinfectious.INTRODUCTIONCoronavirusesarefoundinmanyanima
4、lspeciesincludingmanandareassociatedwithagreatvarietyofdiseases.Theycauserespiratoryandentericdiseasesinhumans,bronchitisinbirds,transmissiblegastroenteritisandencephalitisinpigs,diarrhoeaincalvesanddogs,peritonitisincatsandbothdemyelinatingencephalitisandhepatiti
5、sinrodents(McIntosh,I974;Tyrrelletal.1975).Despitethisgreatvarietyofdiseasesandwidespreadoccurrenceinnature,littlebiochemicalandbiologicalinformationaboutthesevirusesisavailableandevidenceforincludingthesevirusesintoonegroupismainlybasedonmorphologicalandserologic
6、alcharacteristics.InformationoncoronavirusgenomicRNAisavailableforavianbronchitisvirus,twoporcinestrains,onemousestrainandonehumancoronavirus.Thesedataarepartlycon-troversialandincomplete.ThegenomicRNAofavianbronchitisvirus(IBV)consistsofasinglestrandedRNA,whichis
7、polyadenylatedandinfectious(Lomniczi,I977;Schochet-manetal.I977).Evidenceforpolyadenylationwasalsorecentlyreportedforamousehepa-titisvirus(Yogoetal.I977).Estimatesofthemol.wt.ofgenomicRNAwerepublishedforhumanandavianstrains,andvariedbetween5"5to9xio6(Tannock,I973;
