3、-Verlag2003Summary.Porcinerespiratorycoronavirus(PRCV),aspike(S)genedeletionmutantofTransmissiblegastroenteritisvirus(TGEV),causesmildorsubclinicalrespiratoryinfectionsinpigs.ThesheddingofPRCV/TGEVwasstudiedatdifferentdayspost-arrivalinfecalandnasalswabsfromPRCV/TG
4、EVseroneg-ativesentinelpigsintroducedintoaPRCVseropositiveherdwithquestionableTGEVserologyanddiarrhea.NasalsheddingofPRCVwasdetectedin57%and63%ofsamplesbynested-RT-PCRandcellcultureimmunofluorescence(CCIF),respectively.HoweverfecalsheddingofPRCVwasdetectedin37%ofthe
5、samplesbynested-RT-PCRand19%byCCIF.Fourrespiratoryand5fecalPRCVstrainswereisolatedinswinetesticlecellsincludingnasal/fecalPRCVpairs(isolatedatthesametime)from3pigs.Comparisonofnasal/fecalPRCVpairsfromindividualpigsrevealeddifferentdeletionsinthespike(S)gene(648or68
6、1nt)in2pairsandaconsistentchangeinnt790/791(aaTtoV)forallpairs.Inpreliminarystudies,inoculationofgnotobioticpigswitheachplaque-purifiedpairofthenasalandfecalPRCVisolates,revealednoclinicaldiseasebutdifferenttropisms.Thenasalisolatewasshedbothnasallyandinfeces,butthe
7、fecalisolatewasshedonlymarginallyinfeces,andnotnasally.Ourresultsshowthatnested-RT-PCRwasassensitiveasCCIFforPRCVdetectioninnasalswabs,butwasmoresensitivethanCCIFforPRCVdetectioninfecalsamples;alternativelyPRCVshedinfeceswasmorelabilewithlossofinfectivity.TheS-gene
8、sequencedifferencesfoundbetweenthefecalandrespiratoryPRCVisolatesmayinfluencetheirtissuetropism.ThesenewPRCVisolatesshouldbeusefultounderstandthem
