3、isprocessarepoorlyunderstood・Theaimofthisstudywastoexploretheroleofmitogen-activatedproteinkinasekinase6(MKK6)intheHMGBlexpressioninpulmonaryalveolarepithelialcellsinducedbymechanicalstretch.Methods:Pulmonaryalveolarepithelialcells(A549cellline)weredividedinto5groupsbasedonadenoviralinfe

4、ction,includingcontrolgroup,emptyadenovirusvectorgroup,wild-typeMKK6group,constitutivelyactivemutantMKK6(E)group,anddominant-negativemutantMKK6(A)group.Eachgroupwasthensubdividedintostretchedandunstretchedgroups.Theexpressionoftransfectedgeneswasdetectedbyfluorescencemicroscopyandwestern

5、blotting.MKK6activitywasmeasuredusingakinaseactivityassay.TheexpressionlevelsofHMGBlmRNAandproteinweremeasuredbyreversetranscriptionpolymerasechainreaction(RT-PCR)andwesternblotting,respectively.CytokineswereinvestigatedusingtheLiquiChipsystem.Results:Mechanicalstretchsignificantlyenhanc

6、edMKK6activityandHMGBlexpressioninA549cells.TransfectionwithadenoviralMKK6(E)alsoledtoastatisticallysignificantincreaseinHMGBlexpressionlevel,whereastheintroductionofMKK6(A)interferedwithstretch-inducedHMGBlexpression.WealsofoundthatthelevelofHMGBlexpressionwaspositivelycorrelatedwithcyt
