3、calmapsarecrucialtoolsforgenomesequencing,genemappingandcloning,geneticimprovementandselectivebreeding.Inthisstudy,wehavedevelopedagenome-wide,BAC-basedphysicalmapforthespecies.Atotalof81,408clonesfromtwoBAClibrariesofthescallopwerefingerprintedusinganABI3130xlGeneticAnalyzerandafing
4、erprintingkitdevelopedinourlaboratory.Afterdataprocessing,63,641(,5.86genomecoverage)fingerprintswerevalidatedandusedinthephysicalmapassembly.Atotalof3,696contigswereassembledforthephysicalmap.Eachcontigcontainedanaverageof10.0clones,withanaveragephysicalsizeof490kb.Thecombinedtotalp
5、hysicalsizeofallcontigswas1.81Gb,equivalenttoapproximately1.5foldofthescallophaploidgenome.Atotalof10,587BACendsequences(BESs)and167markerswereintegratedintothephysicalmap.Weevaluatedthephysicalmapbyovergohybridization,BAC-FISH(fluorescenceinsituhybridization),contigBACpoolscreeninga
6、ndsourceBAClibraryscreening.Theresultshaveprovidedevidenceofthehighreliabilityofthecontigphysicalmap.Thisisthefirstphysicalmapinmollusc;therefore,itprovidesanimportantplatformforadvancedresearchofgenomicsandgenetics,andmappingofgenesandQTLofeconomicalimportance,thusfacilitatingthegen
