2、niversity,Xi’an,Shaanxi710033,ChinaAbstract:Theauthorsdescribeheretheproceduresforusingthegelatinhalf-embeddingmethodtoobtainthinspinalcordsliceswithattacheddorsalrootsandperformingvisuallyguidedwhole-cellpatch-clamprecordingofpostsynapticcurrentsevokedbyprimaryafferentfibersinratspinaldorsalh
3、orn.Asegmentofspinalcordwithattacheddorsalrootswaspreparedandhalf-embeddedinanagarblockwith20%(w/v)gelatin.Thinspinalcordsliceswithattacheddorsalrootswereobtainedwithavibratomeandwhole-cellpatch-clampconfigurationwasestablishedundertheinfraredobservation.Attheholdingpotentialof70mV,spontaneous
4、excitatorypostsynapticcurrents(EPSCs)anddorsalrootstimulation-evokedEPSCswererecordedasinwardcurrents.Accordingtotheconductionvelocityofafferentfibersandstimulusthreshold,evokedEPSCsthataremediatedbyA-likeorC-likefibersweredistinguished.Attheholdingpotentialof0mV,spontaneousinhibitorypostsynap
5、ticcurrents(IPSCs)anddorsalrootstimulation-evokedIPSCswererecordedasoutwardcurrents.Using5µmol/Lstrychnineor20µmol/Lbicuculline,GABAergicorglycinergicevokedIPSCscouldbeisolated.Usingvisualpatch-clampmethodsynaptictransmissioncanbeaccuratelyassessedbymeasuringpostsynapticcurrentsofthedorsalhorn
6、neurons.Moreimportantly,withtheaidofinfraredobservation,theincidenceoffailuretoestablishaclampconfigurationcanbegreatlyreducedanditbecomeseasiertomakerecordingsfromtheneuronsindeepdorsalhornlaminae.Thus,thepresentresearchapproachaneffectiveapproachtostudythemodulationofprimaryafferentsynaptict
