1、摘要背景与目的：肿瘤的血管发生与肿瘤的生长关系密切。在肿瘤血管发生的分子机制的研究中，DLL4的作用日益受到关注。研究认为，肿瘤血管内皮细胞高表达DLL4，DLL4可能在血管的生长过程中调节着血管内皮细胞的分化增殖，从而达到控制血管生长的作用。本实验拟检测血管内皮细胞是否表达DLL4以及DLL4的两个下游基因HEYl和HEY2。研究DLL4对血管内皮细胞增殖的影响，探索DLL4在肿瘤血管发生和生长中的作用。材料与方法：培养人微血管内皮细胞(HMEC)，MTT法检测HMEC细胞增殖，提取HMEC细胞总RNA，用RT

2、—PCR的方法检测HMEC细胞DLL4mRNA、HEYlmRNA、HEY2mRNA的表达，采用RNA干扰方法观察HMEC细胞DLL4mRNA表达，对结果进行统计学分析。结果：RT-PCR结果显示，HMEC细胞表达DLL4mRNA、HEYlmRNA和HEY2mRNA；DLL4RNA干扰结果显示，lOOnmol／LsiRNADLL4一duplex组和200nmol／LsiRNADLL4一duplex组HMEC细胞DLL4mRNA的表达明显下调，MTT的结果显示lOOnmol／LsiRNADLL4一duplex组和20
