1、上海交通大学硕士学位论文常染色体显性遗传性帕金森病LRRK2基因突变及临床特征分析姓名:张婷申请学位级别:硕士专业:神经病学指导教师:陈生弟20080501上海交通大学医学院01级七年制硕士学位论文森病临床表现,在发病年龄、病程、首发症状、UPDRS评分,对左旋多巴治疗的反应方面与散发性PD患者无明显差异。在本实验中对18位先证者基因筛查,共发现了28个单核苷酸多态,其中12个位于外显子区域,16个位于内含子区域。发现了两个未报道过的外显子单核苷酸多态位点c.851A>G(Asn284Ser)和c.1432G>T(Asp478Tyr)以及6个未报道过的内含子
2、单核苷酸多态位点。Gly2385Arg等位基因突变频率为6.25%,并具有建立者效应。早发型女性PD患者较男性患者更易携带有Gly2385Arg突变。突变携带者与非携带者在临床表现上无明显差异。结论:中国大陆PD人群具有LRRK2基因突变。LRRK2基因Gly2385Arg是中国大陆PD人群种族特异性的危险因素。中国大陆常染色体显性遗传PD患者与散发性PD患者临床表现方面无明显差异。关键词:帕金森病,常染色体显性遗传,LRRK2基因,单核苷酸多态,临床特征II上海交通大学医学院01级七年制硕士学位论文THEMUTATIONANALYSISOFLRRK2GEN
