3、.11,2013;CrosscheckedMay20,2014Abstract:Backgroundandobjective:Ithasbeenshownthatmacrophagesplayanimportantroleinthedevelopmentofsevereacutepancreatitis(SAP),andeventuallyleadtomultipleorganfailure(MOF).Clodronate-liposomeselec-tivelydepletedmacrophages.Thisstudywastoinvestigatetheroleofrena

4、lmacrophageinfiltrationinacuterenalinjuryinratswithSAPandtoevaluatethepotentialofsuperparamagneticironoxide(SPIO)-enhancedmagneticresonanceimaging(MRI)fordiagnosis.Methods:SuperparamagneticFe3O4nanoparticleswerepreparedbychemicalcoprecipitation.SPIO-liposomesandSPIO-clodronate-liposomeswerep

5、reparedbythethinfilmmethod.SAPmodelswerepreparedbyinjectionofsodiumtaurocholateintothesubcapsularspaceofratpancreas.Sprague-Dawleyratswererandomlydividedintoacontrolgroup,SAPplusSPIO-liposome(P)group,andSAPplusSPIO-clodronate-containingliposome(T)group.KidneyinjurywasevaluatedbyT2-weightedMR

6、Iscan.Thelevelsofserumamylase(SAM),bloodureanitrogen(BUN),andserumcreatinine(SCr)weremeasuredbyanautomatedenzymaticmethod.Serumtumornecrosisfactor-α(TNF-α)wasmeasuredbyenzyme-linkedimmunosorbentassay(ELISA).Pathologicalchangesinthepancreasandkidneywereobservedusinghematoxylinandeosin(H&E)sta

7、ining,whilecellapoptosiswasdetectedwithterminaldeoxynucleotidyltransferasedUTPnickendlabeling(TUNEL)staining.Inaddition,themacrophagemarkers(CD68)oftherenaltissueweredetectedwithimmunohistochemistry.Results:Thepathologicalchangesinthepancreasandkid
