4、d-Loxp-EGFP)和TG(zC-myb:EGFP),通过实时跟踪观察,从而动态观测原始造血干细胞和定向造血干细胞的迁移过程,以期了解斑马鱼原始造血和定向造血的关系,为进一步研究斑马鱼血液系统发育提供可能的理论依据。方法分别收集红色荧光蛋白(DsRed)标记原始造血干细胞的转基因系TG(zLmo2:Loxp-DsRed-Loxp-EGFP)和绿色荧光蛋白(EGFP)标记定向造血干细胞的转基因斑马鱼系TG(zC-myb:EGFP)自交和侧交的斑马鱼胚胎,于12hpf(hourspost-fertilization,hpf)去卵膜,进行局部麻

5、醉,利用激光共聚焦显微镜对全胚胎进行连续实时的动态扫描,从12hpf开始观察,直至48hpf,期间每隔两小时拍摄一次进行追踪观察。结果(1)红色荧光蛋白(DsRed)标记原始造血干细胞的转基因系TG(zLmo2:Loxp-DsRed-Loxp-EGFP)自交后,观察胚胎发+现lmo2的细胞在14hpf开始表达红色荧光,主要表达的部位是在斑马鱼胚胎的头部中胚层(anteriorparaxialmesoderm,ALM),随后在中间细胞群(intermediate+cellmass,ICM)表达;18-22hpf发现红色荧光标记的lmo2的细胞在

6、ICM区表达逐渐增强,24hpf达到高峰,30hpf在尾部造血组织(caudalhematopoietictissue,CHT)开始表达,持续至48hpf。(2)绿色荧光蛋白(EGFP)标记定向造血干细胞的转基因斑马鱼系TG(zC-myb:EGFP)自交后,观察绿色荧光标记的+c-myb的细胞,24hpf开始表达,最初表达的部位是PBI(posteriorbloodisland,PBI)区,随后在28hpf表达逐渐增强,并沿着AGM区向原肾导管迁移,28hpf在PBI区和AGM区的表达逐渐增强,持续到36hpf;此外发现在28hpf斑马鱼+的

7、头部和胸腺有少量c-myb细胞的存在;32-36hpfPBI区、AGM区和头部的++c-myb细胞逐渐增多;42-48hpf在斑马鱼头部c-myb细胞逐渐减少(3)TG(zLmo2:Loxp-DsRed-Loxp-EGFP:TG(zC-myb:EGFP)侧交的斑马鱼胚胎,观++察发现受精后24小时可以看到PBI区有少量的又红又绿的lmo2和c-myb的双+++阳性细胞和红色的lmo2的细胞;26hpfPBI区lmo2和c-myb双阳性细胞逐渐++增多,28hpflmo2和c-myb双阳性细胞在PBI区逐渐增多并向主动脉-性腺-3贵阳医学院20

8、13届硕士研究生论文++中肾(aorta-gonad-mesonephros,AGM)迁移,30-38hpflmo2和c-myb双阳性++细胞在尾部PBI区逐渐增多
