1、青岛大学硕士学位论文摘要目的:探讨MicroPuNA-i6通过调节靶基因CCNDI及蛋白表达产物CyclinDl(细胞周期相关蛋白D1),参与上皮性卵巢癌耐药细胞周期调控进程,并研究其与顺铂耐药的关系。方法:MicroRNA-16成体模拟物(mimics)及其阴性对照片段(negativecontrolNC)以脂质体Lipofectamine2000为载体成功转染于人卵巢癌细胞顺铂耐药株SKOV3/DDP细胞;实时荧光定量PCR检测各实验组细胞中miR一16的相对表达量;四甲基偶氮唑蓝(MTT)比色法检测耐药细胞株耐药系数及转染前后细胞顺铂的半数抑制浓度;流式细
2、胞仪检测转染前后各组细胞细胞周期变化;Western—blot蛋白印迹法检测细胞周期相关蛋白CyclinDl蛋白表达量的变化。应用SPSSl7.0进行统计学分析,采用One-wayANOVA单因素方差分析,两组组间比较均采用t检验,尺O.05为差别有显著性。结果:SKOV3/DDP细胞株的耐药系数=4.90;SKOV3/DDP细胞中miR一16表达水平较SKOV3细胞明显降低,差异有显著性意义(P=0.000),转染miR-16mimics后SKOV3/DDP细胞中miR一16水平较转染前明显升高(P=0.001);SKOV3/DDP细胞转染miR一16mimi
